Cell-type-resolved quantitative proteomics map of interferon response against SARS-CoV-2

Summary The commonly used laboratory cell lines are the first line of experimental models to study pathogenicity and performing antiviral assays for emerging viruses. Here, we assessed tropism cytopathogenicity Swedish isolate SARS-CoV-2 in six different human lines, compared their growth characteristics, performed quantitative proteomics susceptible lines. Overall, Calu-3, Caco2, Huh7, 293FT showed a high-to-moderate level susceptibility SARS-CoV-2. In Caco2 cells, virus can achieve high titers absence any prominent cytopathic effect. protein abundance profile during infection revealed cell-type-specific regulation cellular pathways. Type-I interferon signaling was identified as common dysregulated response Huh7 cells. Together, our data show cell-type specific variability cytopathogenicity, susceptibility, provide important clues guide future studies.
